🔬The Polysaccharide Utilization Loci encoded DUF1735 likely functions as membrane-bound spacer for Carbohydrate Active enZymes! A. Lucie Gaenssle, Salvador Bertran Llorens, Tjaard Pijning, Edita Jurak, and I made significant strides in unlocking the function of the Domain of Unknown Function 1735 (DUF1735). Data generated with our Domain and Vicinity Analyzer programs, identified DUF1735 as N-termini of multimodular proteins featuring C-termini like Laminin_G_3, F5/8-typeC, or GH domains, with an Ig-like β-sandwich fold. A function as a membrane-bound spacer for varying C-termini, potentially involved in PUL-mediated carbohydrate utilization, is proposed. In a medium containing esculin, bacteria that produce β-glucosidase can hydrolyze esculin to esculetin, which then reacts with ferric ions in the medium to produce a dark brown or black color.
