High importance and widespread use of Fsp3 and FCstereo indicators in medicinal chemistry and their link to biologically relevant compound features is well known. However, they do not comprehensively express molecular topology, and they often do not match the chemical intuition of complexity. A new empirical scoring system termed the spacial score (SPS) can possibly overcome the existing limitation as it builds and improves upon the individual Fsp3 and FCstereo indicators by uniformly expressing spacial complexity on a highly granular scale for convenient ranking of and comparison between molecules. Furthermore, the SPS is an easy-to-calculate index that does not require information about the 3D conformation of a molecule and, when normalized, correlates to biologically relevant properties, including selectivity and potency. In addition, SPS can be used in the context of synthesis planning and analysis by directly comparing different chemical transformations and their impact on spacial arrangements and molecular complexity in the resulting product's molecular structures.

The chemical stability of all active drugs was evaluated by Ultra-High-Performance Liquid Chromatography tandem mass spectrometry (UHPLC/MS/MS). All samples were analyzed in triplicate at room temperature under normal fluorescent light at 0h, 1h, 6h, 12h, and 24 h. According to the Official Pharmacopoeia of the Italian Republic, the degradation of a molecule that exceeds more than 10% is considered unacceptable. Happy Wednesday all! Check out this recent ACS paper by Sarott et al., "Chemical Specification of E3 Ubiquitin Ligase Engagement by Cysteine-Reactive Chemistry." The authors note "Recently, the attachment of cysteine-reactive chemical groups to pre-existing small molecule inhibitors has been shown to drive specific target degradation. We demonstrate here that different cysteine-reactive groups can specify target degradation via distinct ubiquitin ligases. . . finding that nearly identical compounds can engage multiple ubiquitination pathways suggests that targeting cellular pathways that search for and eliminate chemically reactive proteins is a feasible avenue for converting existing small molecule drugs into protein degrader molecules."
