The field of de novo Protein generation is getting very impressive in Just Two years of developments!!! EvoDiff now enables to generate in minutes (with GPUs) protein (or peptide) sequences that do not necessary met the requirements of a predicted fold, as this system uses diffusion models to learn not how to design only folded proteins from the few hundreds thousand Protein Data Bank structures, but how natural sequences can perform their function, folded or not, directly from the full protein Universe (> 300 million sequences). Apart from a superior flexibility than previous approaches in the task of DE-NOVO Protein sequences generation, it can also recieve further constraints from a user-supplied multiple sequence alignment to generate artificial proteins by sampling from the diversity of Natural Protein families. With some more developments, the classic Protein Discovery approaches from Natural diversity will be essentially rule out as old and too much time-consuming, while being replaced with a human-conditioned sampling of artificial Protein sequences that have an high likelihood to possess the property we need. I am so excited to share EvoDiff, diffusion models trained on evolutionary-scale data that operate in the protein sequence space. We show that EvoDiff generates high-fidelity, diverse, and structurally-plausible proteins that fully cover natural sequence and functional space. We also show that EvoDiff can be used to scaffold structural motifs, demonstrating that sequence may be all you need to controllably design new proteins Our preprint is live, the code is open source, and we are excited to see how researchers will use it!
