Nowadays, they are a very useful reproduction, reagent, and device in various scientific disciplines, including medicine, chemistry, biochemistry, colloid science, biology, physics, biophysics, mathematics and theoretical. After the initial discoveries liposomes have made their way to the market. Among numerous brilliant new drug delivery systems developed, liposomes characterize an advanced technology to deliver active molecules to the site of action, and at present, several formulations are in clinical use. Research on liposome technology has progressed from conventional vesicles to ‘second-generation liposomes’, in which long-circulating liposomes are obtained by modulating the lipid composition, size, and charge of the vesicle. Liposomes with modified surfaces have also been developed using several molecules, such as glycolipids or sialic acid. Bridging for therapeutic drug monitoring: The gold standard biological matrix for measuring drug concentration in blood is plasma, but plasma has drawbacks that can lead to underrepresenting drug concentration in plasma-based drug studies. Measuring the drug in blood that contains cells is vital to get a representative drug concentration. With improvements in analytical technology, it is possible to measure these drugs from whole blood #microsamples. But if an assay was initially validated in venous plasma, then clinical bridging needs to be conducted in advance to understand what biases are seen when transitioning an assay to whole blood. Researchers at UCB Biopharma S/A did this using #Mitra w/ #VAMS remote #microsampling. Diagnostics led continuous learning dosing models in clinical practice and at home therapeutics dosing 🔸 SUMMARY In this note we discuss combing therapeutics, diagnostics and quantitative system pharmacology modelling to provide a patient personalized precision dosing strategy, for achieving therapeutic efficacy whilst reducing toxicology.   🔸 BACKGROUND Modelling in the biopharma industry has been a part of the toolset for over 30 years, with Silicon Graphics machines embedded in Medicinal Chemistry Departments by the mid-nineties. Back then, modelling was narrow and specific, focused on small molecule and binding site interactions. Nowadays, modelling is also used at a systems level, such as in Quantitative Systems Pharmacology (QSP), which models the dynamic interaction between drugs and biological systems. There is also the related subject of Quantitative Systems Toxicology (QST), which aims to reduce the toxicological impact of drug therapy rather than focusing solely on broader pharmacological activity.
