If you are attending the #acsfall2023 meeting consider tuning into my presentation entitled, "Discovery of benzothiophene difluoromethyl phosphonates (DFMP) as potent PTPN1/2-dual inhibitors through structure-based drug design (SBDD)". Historical challenges with drugging protein tyrosine phosphatases (PTPs) have earned this class of targets a reputation for being “undruggable”. Indeed, the team encountered several challenges on this program resulting from the physchem properties of the negatively charged difluoromethylphosphonate that put us squarely beyond “Rule of 5” chemical space.
